The only way to kill Samedi is by shooting/stabbing him in the eye. None. + Samedi's ability is impenetrable skin, so his eyes shouldn't also be impenetrable.
- Not if he closes his eyes, which then would be protected by his eyelids.

- If someone manages to stab would him from inside his mouth, he would be killed.

- In a deleted scene, Sylar acquired this ability from another person by reaching said person's brain through a soft spot - the mouth.
